誘電泳動インピーダンス計測法による
迅速かつ簡便な細菌・ウイルス・DNA検出
誘電泳動(Dielectrophoresis, DEP)とは外部電界によって分極した誘電体粒子に作用する静電気力によって、粒子が高電界または低電界領域に駆動される動電現象のことです。直流電界を用いる電気泳動と異なり、誘電泳動では交流電界を用いることができるので、水に懸濁した細胞などの生体物質を水の電気分解を抑制しつつ操作することができます。更に、細胞の誘電特性の周波数依存性を利用して、細胞の分離同定などに応用することもできます。このような特徴を生かして、誘電泳動をバイオテクノロジーに応用する試みが近年活発になっています。
1996年,我が国では病原性大腸菌O157による集団食中毒が大きな社会問題となりました。このような問題を解決するため、簡易・迅速・低コストでかつ自動化が可能な細菌検査法が強く求められています。また,食品の品質管理以外に,医療診断や環境アセスメントなどの分野でも簡易かつ迅速な細菌検出法に関するニーズが存在します。私達は、誘電泳動と電気インピーダンス計測を組み合わせた新しいバクテリア検出法である”DEPIM”を開発しました(特許出願中)。DEPIMでは、誘電泳動により細菌を集積型マイクロ電極上で濃縮し、その時の電極間インピーダンスの変化から細菌濃度を定量的に推定できます。
DEPIMは培養法などの従来の細菌検出法に比べ簡便かつ迅速であるという特徴を有しています。また、細菌物性値の活性依存性や抗原抗体反応を利用することにより、菌活性や菌種による選択的検出も可能です。更にパルス電界によって細菌の細胞膜を破壊して細胞質に含まれる電解質イオンを流出させることにより、1ccあたり100個程度の細菌を3時間以内に定量的に検出することができます。現在、産学共同研究により、食品衛生管理や口腔衛生管理に応用可能なDEPIM装置の実用化にも取り組んでいます。
